I think the love Dean feels for Cas actually defies gender. So yeah, in a way Cas’ vessel doesn’t matter.

BUT. I ALSO think Dean has been suppressing his sexuality for a long long time. So the fact Cas is in a male vessel is INCREDIBLY BENEFICIAL to Dean’s development. Because yes, he DOES love Cas regardless of gender, and that’s gonna HELP him embrace the fact that actually the male body is something he’s always had an attraction to, so getting physically close to Cas will get him Cas and it will also allow him to admit to and make peace with his sexuality.
